DSIP is a naturally occurring nonapeptide discovered in 1974 that promotes delta wave sleep, reduces stress, and modulates multiple neurotransmitter systems including GABA and endogenous opioids. It has a molecular formula C35H48N10O15, mass 848.8 Da. Its published amino-acid sequence is Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu. What it's studied for
Investigation of DSIP to date is primarily preclinical, with the majority of citations concentrated in animal models and in-vitro assays rather than randomized controlled human trials. Published work referencing DSIP spans Sleep architecture studies: EEG studies in rabbits, rats, and limited human cohorts show DSIP increases delta wave (slow-wave) sleep duration without suppressing REM sleep. Unlike benzodiazepines, it enhances physiological sleep patterns rather than altering them, Chronic insomnia trials: European clinical studies (primarily 1980s-1990s) in patients with chronic insomnia reported improved sleep onset latency, reduced nocturnal awakenings, and enhanced subjective sleep quality. Schneider-Helmert (1983) reported significant improvements in 15 of 16 patients with psychophysiological insomnia, and Stress and HPA axis: DSIP reduces ACTH and cortisol secretion, suggesting anxiolytic and stress-protective properties. Research in alcohol and opioid withdrawal contexts showed DSIP reduced withdrawal symptom severity and normalized stress hormone levels. Proposed mechanisms in the literature include GABA system enhancement and NMDA receptor interaction.
